Gebelikte kontrol edilemeyen konvülsif nöbetler yavruların gelişimi üzerinde
olumsuz etkilere sahiptir. Bu çalışmada, gebelerde kullanımı daha güvenli bir
antikonvülsan olan lamotrijin ile gelişimsel nörotoksisite bakımından daha
fazla risk taşıyan fenobarbital‟in deney hayvanlarının motor ve öğrenme
becerileri üzerindeki etkilerinin karşılaştırılması amaçlanmıştır. Cerebellum
yeni motor becerilerin öğrenilmesinde kritik bir öneme sahip olduğu ve
çıktıların kaynağı olan Purkinje nöronlarının gelişimi sıçanlarda embriyonik
(E) 14. günden itibaren başladığı için ilaç uygulamaları E14-E21 arasında
yapılmıştır. Deney gruplarına (n=7 her grup ve cinsiyet için) oral gavaj yoluyla
lamotrijin ve fenobarbital (20 mg/kg), kontrol grubuna da serum fizyolojik
uygulanmıştır. Yavrular postnatal 40. günden itibaren rota-rod, yükseltilmiş
artı labirent, spontan lokomotor aktivite, açık alan, beceri gerektiren kavrama
ve grip testlerine tabi tutulduktan sonra sakrifiye edilmiştir. Cerebellum‟dan
sistematik randomize örnekleme yöntemiyle alınan kesitler üzerinde
nöronların ve liflerin bulunduğu tabakaların hacimsel oranları ile beyaz
cevherdeki myelin dansiteleri ölçülmüştür. Aktivitemetrede lamotrijin
uygulanan yavrularda cinsiyetten bağımsız olarak, fenobarbital
uygulananlarda ise dişilerde olmak üzere kontrollere göre anlamlı bir düşüş
gözlenmiştir. Rota-rod testinde de fenobarbital uygulanan erkeklerin motor
becerilerinde anlamlı bir bozukluk saptanmıştır. Birim uzunluk başına düşen
Purkinje hücre sayısı fenobarbital grubunda diğer gruplara göre anlamlı
derecede düşük iken; kortikal tabakalarının hacimsel oranlarında ve beyaz
cevherdeki miyelin dansitesinde anlamlı bir farklılık gözlenmemiştir.
Uncontrolled convulsive seizures during pregnancy have negative effects on
the development of offspring. In this study, we aimed to compare the effects
of lamotrijin, which is a safer anticonvulsant drug in pregnant women, and
phenobarbital, which has a higher risk of developmental neurotoxicity, on the
motor and learning skills of the experimental animals. Since the cerebellum
has critical importance in learning new motor skills and development of the
Purkinje neurons, the main source of outputs, begin on the embryonic day
(E) 14 in rats, drug applications were carried out between E14-E21.
Experimental groups (n = 7 for each group and gender) were administered
lamotrijin and phenobarbital (20 mg/kg) by oral gavage and saline was
administered to the control group. From the postnatal 40th day onwards, the
offspring were subjected to rota-rod, elevated plus maze, spontaneous
locomotor activity, open field, grip and fine-skill required grip tests, and then
sacrificed. Systematic randomized sampling method were applied on
sections of the cerebellum to estimate the volumetric ratios of the cerebellar
layers in which neurons and fibers are located, and then myelin densities
were measured in the white matter. In activitymeter, a significant decrease
was observed in the offspring treated with lamotrijin, and in female offspring
treated with phenobarbital. Rota-rod test also showed a significant decrease
in the motor skills of males treated with phenobarbital. The number of
Purkinje cells per unit length was significantly lower in the phenobarbital
group than in the other groups. No significant difference was observed in the
volume proportions of the cortical layers and myelin density in the white
matter
