Kemoterapide, siklofosfamid (CP) gibi yüksek doz alkilleyici ajanlar, çoklu
organ toksisitesine neden olmaktadır. Testiküler hasar da yaygın olarak görülmesine
rağmen, antikanser ilaçların bu yan etkisi diğer yan etkilere göre daha az araştırılmıştır.
Bizim çalışmamızda da sıklıkla kullanılan bir antikanser ilaç olan CP’ nin testiküler
toksisiteye karşı, Hipericum triquetrifolium Turra (HT, Kantaron) bitki ekstresinin
hücre koruyucu etkisi araştırıldı. Bu amaçla Wistar cinsi sağlıklı, 3-4 aylık, 200±20 g,
erkek albino sıçanlar her grupta 7 hayvan olacak şekilde 9 gruba ayrıldı (kontrol/ 150
mg/kg CP/ 25, 50, 100 µg/ml HT grupları/ CP+25, 50, 100 µg/dl HT grupları ve 0.2 ml
DMSO). Tüm enjeksiyonlar intraperitonal olarak yapıldı. CP hariç diğer kimyasallar 6
gün boyunca her gün verildi. CP’ li gruplarda, CP, 6. gün tek doz olarak verildi ve 7.
gün ketamin/ksilazin anestezisi altında kan ve doku örnekleri alınarak hayvanlar
öldürüldü. Alınan örnekler biyokimyasal, histopatolojik, immünohistokimyasal (kaspaz
3, Bcl-2, Bax) ve Lazer Taramalı Konfokal Mikroskop (LTKM) ile incelendi. HT 'nin
antioksidan etkilerini saptamak için biyokimyasal parametrelerden malondialdehid
(MDA), glutatyon (GSH), süperoksitdismutaz (SOD), katalaz (CAT), glutatyon
peroksidaz (GPx), toplam antioksidan seviye (TAS), toplam oksidan seviye (TOS),
oksidatif stres indeksi (OSİ) düzeylerine bakıldı. CP verilen gruplarda MDA ve TOS
’nin artması ile GSH ve TAS’nin azalması CP nedenli oksidatif hasarı gösterdi.
Histopatolojik ve İmmünohistokimyasal olarak dokuların boyanması, CP nedenli
testiküler hasarı açık bir şekilde gösterdi. Deneysel sonuçlarımız, sıçanlarda HT'nin
metanol ekstresinin 100 mg/kg’lık dozunun diğer dozlarına göre daha etkili ve CP’ye
karşı koruyucu olduğunu göstermektedir. Bu çalışma ESOGÜ /HADYEK izni ile

High-dose alkylating agents such as cyclophosphamide (CP) in chemotherapy
cause multiple organ toxicity. Although testicular injury is also common, this side effect
of anticancer drugs has been less studied than other side effects. Therefore, the cell
protective effect of Hypericum triquetrifolium Turra (HT) extract against testicular
toxicity was investigated. With this in mind, healthy male albino Wistar rats, aged 3-4
months weighing 200±20 g, were divided into 9 groups, each of them containing 7
members (Control, 150 mg/kg CP/ 25, 50, 100 µg/ml HT Groups, CP+25, 50, 100 µg/dl
HT Groups and 0.2 ml DMSO). All the injections were applied intraperitonally. Except
for CP, all the other chemicals were given for 6 consecutive days. In the CP groups, the
drug was given only one dose a day. On the 7th day, the rats were sacrificed under
ketamine/xylazine anaesthesia before their blood and tissue samples could be taken for
analysis. The samples were then analysed biochemically, histopathologically and
immunohistochemically (caspase 3, Bcl-2, Bax). In order to determine the effects of
antioxidant properties of HT, MDA, GSH, SOD, CAT, TAL, TOL and OSI were
analysed. In the CP group, decreased GSH and TAS with increase in MDA and TOS
showed oxidative damage caused by CP. Caspase 3, Bcl-2, Bac immunoactivities
showed CP-related testicular damage depending on the severity of coloration. Our
experimental results show that the methanol extract of HT is more effective and
protective against CP than other doses of 100 mg/kg in rats. This study was conducted
