Son yıllarda ilaç bulma ve geliştirme çalışmalarında, biyolojik etkinliği bilinen bir
molekülden yola çıkılarak kimyasal veya biyoteknolojik yöntemlerle türevlendirme ile
daha yüksek biyolojik aktiviteye ve daha az yan etkiye sahip ilaçların elde edilmesine
yönelik çalışmalar önem kazanmıştır.
Bu çalışmada farklı biyolojik aktivite gösterdiği bilinen betülin molekülünün
bakteriyel kültürler ile biyotransformasyontepkimeleri sonucu türevlerinin elde edilmesi,
betülin ve elde edilmesi muhtemel türevlerin antimikrobiyal ve A549, HepG2 ile 5RP7
kanser hücrelerine karşı apoptotik etkilerinin ortaya konması amaçlanmıştır. Betülinin 7
farklı bakteri (StaphylococcusaureusATCC 6538, ProteusvulgarisNRRL B-123,
BacillussubtilisNRRL B-4378, StreptomycesgriseolusNRRL B-1062,Escherichia
coliATCC 8739, StaphylococcusaureusATCC 43300 ve BacillusvelezensisNRRL B 14580) ile gerçekleştirilen biyotransformasyon tepkimelerinden saf metabolit üretilemedi.
Antikanser aktivite çalışmalarında, betülin molekülünün A549, HepG2 ve 5RP7 hücre
dizilerine karşı sırasıyla 207.7, 125.0 ve 28.3 µg/mL İK50 değerlerine sahip olduğu
bulunmuştur. SI değerleri ise sırasıyla 30, 50 ve 223 olarak belirlenmiştir. Bu durum
betülinin aktivitesininA549 ve HepG2 hücrelerine karşı seçici olmadığı ve sağlıklı
hücrelere karşı sitotoksisitesinin etkinliğini sınırlandırdığını, ancak 5RP7 (28.33±1.53
µg/mL) hücresine karşı seçici olarak sitotoksik etkinlik gösterdiğini söylemek mümkündür.
Ayrıca bunun etkisinin apoptotik mekanizmaya dayandığı gösterilmiştir
In recent years, research Studies based on derivatisation of a biologically active
molecules through chemical and Biotechnological methods to develop more active drug
ingredients with less side effects have gained importance in finding and developing new drug
canditate molecules studies.
In this study it was aimed at obtaining derivatives of betuline which is known to
exhibit a variety of Biological properties through biotransformation Reactions with bacterial
strains and investigating the antimicrobial activities and apoptotic effects against A549,
HepG2 and 5RP7 cancer cell of betuline and its possible metabolites. Biotransformation
reactions of betuline by 7 bacteria (Staphylococcus aureus ATCC 6538, Proteus vulgaris
NRRL B-123, Bacillus subtilis NRRL B-4378, Streptomyces griseolus NRRL B-1062,
Escherichia coliATCC 8739, Staphylococcus aureus ATCC 43300 and Bacillus velezensis
NRRL B-14580) produced no pure metabolite. In in vitro anticancer activity studies, betuline
was found to exert anticancer activity against A549, HepG2 and 5RP7 cell lines with an IC50
value of 207.7, 125.0 and 28.3 µg/mLwhereas SI values were found to be 30, 50 and 223,
respectively. This indicated that betuline activities against A549 and HepG2 cell lines were
nonselective and limited its cyctotoxic activity against healthy cells but it is possible to say
that it exerted selective activity against 5RP7 cell (28.33±1.53 µg/mL). In addition, it was
shown that its effect is based on apoptotic mechanism
