Bu çalışmanın ilk aşamasında Agaricus bisporus (beyaz suş), Agaricus bisporus
(kestane suşu), Boletus edulis, Cantharellus cibarius, Ganoderma lucidum, Pleurotus
ostreatus ve Trametes versicolor makrofunguslarından ekstrakte edilen polisakkaritlerin in
vitro prebiyotik etkileri araştırılmıştır. Bu amaçla Lactobacillus plantarum, Lactobacillus
acidophilus ve Escherichia coli bakterilerinin % 0,25, % 0,5, % 1,0 ve % 2
konsantrasyonlarında polisakkarit varlığında üremesi 48 saat süre ile mikropetri
kuyularında nefalometrik olarak izlenmiştir. Negatif ve pozitif kontrol grubu olarak sırası
ile glikoz ve inulin kullanılmıştır. En yüksek in vitro prebiyotik aktivite Cantharellus
cibarius polisakkaridinin (CCP) % 0,25 konsantrasyonda elde edilmiştir. Çalışmanın
ikinci aşamasında % 0,25 CCP’nin in vivo koşullardaki prebiyotik etkisi mikrobiyolojik,
biyokimyasal ve histolojik parametreler ile araştırılmıştır. Mikrobiyolojik parametre
olarak, in vivo deneylerden elde edilen bağırsak örneklerinde Lactobacillus spp.,
Bifidobacterium spp., Bacteroides spp. ve Clostridium spp. sayımı yapılmıştır.
Lactobacillus spp. ve Bifidobacterium spp.’nin en fazla üremesi CCP ile beslenen
sıçanlarda olmuştur. En yüksek Bacteroides spp. ve Clostridium spp. üremesinin ise
negatif ve pozitif kontrol grubunda olduğu belirlenmiştir. Elde edilen veriler ışığında
CCP’nin prebiyotik olarak bir potansiyele sahip olduğu belirlenmiştir
In the first step on this study the in vitro prebiotic activity of some mushrooms
(Agaricus bisporus (white), Agaricus bisporus (brown), Boletus edulis, Cantharellus
cibarius, Ganoderma lucidum, Pleurotus ostreatus and Trametes versicolor)
polysaccharides were investigated. For this purpose, the growth of Lactobacillus
plantarum, Lactobacillus acidophilus and Escherichia coli bacteria in presence of 0.25%,
0.5%, 1.0% and 2% concentrations mushroom polysaccharides was monitored
nephometrically in microplate wells during 48 hours. Glucose and inulin were used as
negative and positive control group, respectively. The highest in vitro prebiotic activity
was obtained in the 0.25% dose of Cantharellus cibarius polysaccharide (CCP). In the
second step of the study, the in vivo prebiotic effect of CCP (0,25%) was investigated by
microbiological, biochemical and histological parameters. The colonic Lactobacillus spp.,
Bifidobacterium spp., Bacteroides spp. and Clostridium spp. were counted as
microbiological parameters. The highest colony numbers of Lactobacillus spp. and
Bifidobacterium spp. were obtained in the group of CCP. On the other hand, the highest
Bacteroides spp. and Clostridium spp. colony numbers were counted in negative and
positive control groups. As a result, it has been observed that CCP has a potential as a
prebiotic
