Bu çalıĢmanın amacı, hipersalin ortamlara adapte olmuĢ mikrofungus izolatlarının
antioksidan ve antimikrobiyal aktivitesi açısından taranarak etkin metabolit üreticileri
belirlemek ve üretim koĢullarını istatistiksel tasarım yöntemleriyle ortaya koymaktır.
Ġlk olarak 300 fungal izolat in vitro antioksidan ve antimikrobiyal aktivite açısından
basit yöntemlerle taranmıĢtır. Antioksidan ve antimikrobiyal aktiviteyi değerlendirmek için
DPPH serbest radikal süpürücü aktivite tayin yöntemi ve agar blok difüzyon testi
kullanılmıĢtır.
Aspergillus izolatlarının sıvı kültürde 27°C‟de 7 günlük inkübasyon sonucunda elde
edilen filtratların etil asetat özütlerine antioksidan ve antimikrobiyal aktivite testleri
uygulanarak ikinci defa taranmıĢtır. Bulgular, denenmiĢ diğer cinslere göre daha fazla
oranda Aspergillus ve Penicillum türünün etkili metabolitler ürettiğini destekleyici
yöndedir.
Tarama sonuçlarına göre seçilen CT 4.24 kodlu Aspergillus clavatus izolatının
etkin metabolit üretim koĢulları istatistiksel yöntemlerden sırasıyla Plackett Burman ve
Full faktöriyel tasarım kullanılarak optimize edilmiĢtir. A. clavatus ile metabolit üretiminde
sükroz ve peptonun önemli besiyeri bileĢenleri olduğu, sükrozun denenen 30 ve 50 g/l
konsantrasyonlarının etken madde üretimini olumlu yönde etkilediği belirlenmiĢtir.
Belirlenen optimum koĢullarda yapılan üretim sonucu elde edilen etil asetat özütünün
serbest radikal süpürücü aktivitesi %94,40 (DPPH) ve minimum inhibisyon
konsantrasyonları ise S. aureus ATTC 29213 için 62,5 μg/ml, E. coli ATTC 25922 için
125 μg/ml olarak tespit edilmiĢtir.
Sonuçlar hipersalin ortamlardan izole edilen halotolerant/halofilik funguslar
tarafından üretilen metabolitlerin yeni doğal antioksidan ve antimikrobiyal bileĢiklerin
potansiyel kaynağı olabileceğini ortaya koymaktadır.
The purpose this study is to identify the producers of effective metabolites by
screening antioxidant and antimicrobial activities of microfungi isolates adjusted to
hypersaline media and to reveal production conditions by statistical design methods.
Firstly, 300 fungal isolates were screened in vitro for antioxidant and antimicrobial
activity by simple methods. DPPH free radical scavenging activity determination assay and
agar block diffusion assay were used to evaluate antioxidant and antimicrobial activity.
Aspergillus isolates were screened twice by applying antioxidant and antimicrobial
activity tests on ethyl acetate extracts of filtrates obtained in liquid culture at 27° C for 7
days as a result of incubation. The findings support the more effective metabolites of
Aspergillus and Penicillum species compared to other genus tested.
The effective metabolite production conditions of CT 4.24 coded Aspergillus
clavatus isolates selected by screening results were optimized using statistical methods,
Plackett Burman and Full factorial design, respectively. It has been understood that sucrose
and peptone are important nutrient components in the production of metabolites by A.
clavatus, and sucrose concentrations of 30 and 50 g/l are effective in the production of
active ingredients positively. The minimum inhibitory concentrations of the ethyl acetate
extract obtained from the determined optimum conditions were 62.5 μg / ml for S. aureus
ATTC 29213, and 125 μg / ml for E. coli ATTC 25922.
The results reveal that metabolites produced by halotolerant/halophilic fungi
isolated from hypersaline environments can be a potential source of novel natural
antioxidant and antimicrobial compounds.
