Bu çalışmada, bazı kumarin türevlerinin (3-asetamidokumarin, (3-AC); 3-a(asetonilbenzil)-4-hidroksikumarin ve varfarin, (4-OHC); 4-hidroksi-3-(3-okso-1fenilbütil)kumarin)) asitlik sabitleri (pKa) ve tautomerik denge sabitleri (KT) gibi tanıtıcı fiziksel ve kimyasal parametreler, deneysel ve teorik olarak saptanmıştır. Tautomerik yapılarının kuantum kimyasal hesaplamaları ile incelenmesi sağlanmış ve yapı-etki (QSAR) ilişkilerinin aydınlatılmasına çalışılmıştır. Bu amaçla moleküllerin nötr durumları, proton vermiş anyonları, proton almış halleri ile model moleküller modellenmiştir. Ayrıca, teorik olarak hesaplanan parametreler ile deneysel veriler arasındaki korelasyon araştırılmış ve kabul edilebilecek düzeyde uyum gözlenmiştir. Moleküllerin gaz ve sıvı fazda, fiziksel ve termodinamik özellikleri ile yapısal parametreleri Hartree-Fock (HF) metodu ve Yoğunluk Fonksiyonu Teorisi (DFT/B3LYP) hibrit yaklaşımı ile 6-31G(d) kullanılarak hesaplanmıştır. Sonuçta, uyumun en iyi olduğu durumlar seçilerek proton-verme/alma merkezleri ve mekanizmaları saptanmaya çalışılmıştır. Bulunan asitlik sabitleri pKa değerleri, kullanılarak keto-enol tautomerik denge sabitleri, KT, hesaplanmış ve enol formlarının daha kararlı olduğu gözlenmiştir.
Çalışmamızda ayrıca teorik ve deneysel NMR spektrumları incelenerek moleküllerin yapıları aydınlatılmıştır. Bu amaçla moleküllerin ab initio Hartree-Fock ve yoğunluk fonksiyon metodları (DFT) metodları ve farklı temel setlerde gerçekleştirilen optimizasyon işlemlerinden sonra, 1H ve 13C izotropik kimyasal kayma değerleri Gaussian G03W Programı kullanılarak, atomik orbitalleri içeren GIAO metoduna göre hesaplanmıştır.
In this study, some physical and chemical parameters of two coumarin derivatives (3-acetamidocoumarin (3-AC), 3-a-(acetonylbenzyl)-4-hidroxycoumarin), warfarin, (4-OHC); 4-hydroxy-3-(3-oxo-1-fenylbutyl)coumarin)) such as acidity constants (pKa) and tautomeric equilibrium constants (KT) have been investigated by theoretically and experimentally. The tautomeric structures were determined by using quantum chemical calculations. The obtained data were used to search the relations of Quantitative Structure Activity Relations (QSAR). For this purpose, neutral, deprotonated and protonated forms of these molecules have all been modeled by molecular modeling. Also it has been investigated the correlation between theoretical parameters and experimental data and acceptable results have been observed. The physical, thermodynamic properties and structural parameters of these molecules have been determined by Hartree-Fock and Density Functional Theory calculations at 6-31G(d) basis set. Taking into account of the best fit results, the deprotonation/protonation centers and patterns have been determinated. Using the calculated acidity constants, the equlibrium constants, KT, for keto-enol tautomerizm have been calculated. The evaluation of the tautomeric equlibrium constant values revealed that the enol forms are more stable.The structures of the molecules have been clarified by NMR spectra. For this purpose, after the optimizations which have been done with ab initio Hartree-Fock and DFT methods, 1H and 13C isotropic chemical shift values have been calculated according to GIAO method including atomic orbitals by using Gaussian 03W software programme
