İskemi reperfüzyon hasarı birçok patofizyolojik süreçten meydana gelen karmaşık olaylar dizisidir. Serbest radikaller, tek elektron eksiklikleri nedeniyle başka moleküllerle kolayca elektron alışverişi yapabilir veya onlarla birleşebilirler. Böylece diğer molekülerin yapı ve fonksiyonlarını değiştirebilir, hatta pek çok dokuda hücre hasarı meydana getirebilirler. Antioksidanlar hem direkt hem de dolaylı olarak hücre içerisinde meydan gelen toksik radikal reaksiyonların oluşturduğu oksidatif hasarın olumsuz etkilerine karşı hücreleri koruyan moleküllerdir. İskemi sırasında meydana gelen serbest radikallerin etkisinden korunmak için antioksidan maddeler kullanılabilir. Silimarin ve likopen gibi antioksidan özelliğe sahip maddeler hücre koruyucu özelliğe sahiptirler. Bu çalışmada deney hayvanlarına 45 dakika sıcak iskemiyi takip eden 6 saatlik reperfüzyon süresinde oluşan oksidatif stres kaynaklı hücresel hasarın likopen (2.5, 5 ve 10 mg/kg dozlarda) ve silimarin (50 ve 100 mg/kg dozlarda) ile olası koruyucu rolü araşırıldı. Bu çalışmada; - Kan; nötrofil, lenfosit, toplam lökosit ve eritrosit değerleri, - Serum; kreatinin, üre ve ürik asit değerleri ile alanin amino transferaz, aspartat amino transferaz ve laktat dehidrogenaz enzimleri, - Böbrek malondialdehit miktarı ile hücresel antioksidan enzimler olan katalaz, glutatyon ve süperoksit dismutaz enzimlerindeki önemli değişiklikler araştırıldı. Çalışmadan elde edilen sonuçlar, böbrek iskemi reperfüzyonu sırasında likopen uygulamasının özellikle 2,5 mg/kg’lık dozunda, meydana gelen hücresel hasarı azaltabileceğini göstermiştir.
Ischemia-reperfusion injury is a complex chain of events consisting of many pathophysiological processes. Free radicals, due to a missing electron, can easily transfer electrons to other molecules or combine with them. As a result, they can modify the structure and functions of other molecules; moreover, they may cause cell damage on many tissues. Antioxidants are molecules that protect the cells, both directly and indirectly, against the harmful effects of the oxidative injury caused by toxic radical reactions occurring inside the cell. Antioxidants can be used to protect the cells against the effects of free radicals forming during Ischemia. Antioxidants such as silymarin and lycopene have cell-protection characteristic. This study assessed the potential protective role of lycopene (2.5, 5 and, 10 mg/kg doses) and silymarin (50 and 100 mg/kg doses) on oxidative stress based cell damage occurred as a result of 6-hour reperfusion following 45-minute warm ischemia in test animals. In this study - Blood values (neutrophil, lymphocyte, total leukocyte and erythrocyte), - Serum; creatinin, ure and uric acid values and, alanin aminotransferase, aspartat aminotransferase and lactate dehydrogenase enzymes, - Renal malondialdehyte value and significant changes in the amount of katalase, glutattion and superoxide dismutase enzymes which are cellular antioxidant enzymes were assessed. The results of the study have pointed out that lycopene administration (especially on 2,5 mg/kg dose) can reduce cellular damage occurred during renal ischemiareperfusion.
