Bu çalısma akut strok hastalarında plazminojen aktivatör inhibitör tip-1 (PAI-1)
geni 4G/5G polimorfizmi dagılımı ve plazmada PAI-1 enzim aktivitesini belirlemek ve
aralarında bir iliski olup olmadıgını belirlemek amacıyla yapıldı.
Çalısmada akut strok (iskemik ve hemorajik) tanısı konmus 253 hasta ile kontrol
amaçlı kullanılmak üzere 80 saglıklı kisi olmak üzere toplam 333 kisiden elde edilen
genomik DNA’lar kullanıldı. Genomik DNA’lar venöz kandan tuz yöntemi kullanılarak
elde edildi. DNA’lar 4G ve 5G alellerine özel primerler kullanılarak PCR yöntemi ile
çogaltıldı. PCR ürünleri % 2 agaroz jel elektroforeze tabi tutularak CCD kamera ile
degerlendirildi. Plazma plazminojen aktivatör inhibitör tip-1 enzim aktivitesi ELISA
yöntemiyle ölçüldü. Sonuçlar student t-testi, 2, tek yönlü varyans ve stepwise regresyon
analizleri ile degerlendirildi.
Arastırmada; akut strok hastaları ile kontrol grubunda PAI-1 geni 4G5G genotip
sıklıgı diger genotiplere göre düsük bulundu. PAI-1 enzim aktiviteleri hastalarda
kontrol grubuna göre önemli derecede yüksek olarak belirlendi. Hastalar arasında
4G5G genotipi sıklıgı en az olmasına karsılık bu genotipe sahip bireylerde PAI-1 enzim
aktivitesinin en yüksek oldugu, buna karsılık 4G4G genotip sıklıgı en fazla olmasına
ragmen bu genotipe sahip bireylerde plazma PAI-1 enzim aktivitesinin en düsük oldugu
belirlendi. Plazma PAI-1 enzim aktivitesi artısına homosistein düzeylerinin % 65 pozitif
etkili oldugu belirlendi.
Sonuç olarak, PAI-1 geni 4G/5G genotipleri ile plazma PAI-1 enzim aktivitesi
arasında strok açısından bir iliski saptanmadı. Buna ragmen, PAI-1 geni 4G/5G
genotipleri, plazma PAI-1 enzim aktivitesi ile homosistein düzeyi tesbitinin kisilerin
ileride strok geçirip geçirmeyeceginin belirlenmesinde önemli yardımcı kriterler
olabilecegi belirlendi.
This study was carried out to determine plasminogen activator inhibitor type-1
(PAI-1) gene 4G/5G polymorphism and plasma PAI-1 activity in acute stroke patients,
and to establish whether there is an association between this polymorphism and plasma
activity.
In this study 253 acute stroke patients and 80 healthy subjects as a control of 333
genomic DNA were analysed. Genomic DNA was prepared from peripheral blood by
using salt method. DNA’s were amplified by PCR method using primers spesific for 4G
and 5G alleles. PCR products were seperated by 2 % agarose gel electrophoresis and
visualized by CCD camera. PAI-1 enzyme activities were measured by ELISA method.
The results were evaluated statistically by using student t-test, 2 test, one way analysis
of variance and stepwise regression analysis.
In this study; frequency of PAI-1 gene 4G5G genotype was found low both in
patients and controls. PAI-1 enzyme activities were increased significantly in acute
stroke patients according to controls. While 4G5G genotype frequencies were lowest,
patients carrying this allele had highest plasma PAI-1 enzyme activity. Although 4G4G
genotype frequencies were highest, patients carrying this allele had lowest plasma PAI-1
enzyme activities. Homocystein levels had 65% positive effect plasma PAI-1 enzyme
activities.
Consequently; any association between PAI-1 gene 4G/5G genotypes and plasma
PAI-1 enzyme activities according to stroke is determined. Although, determining PAI-1
gene 4G/5G genotypes, plasma PAI-1 enzyme activities and homocystein levels is defined
as important accessary criterias would determine future stroke risk.
