Invaziv meme karsinomlarının progresyonunda fascin’in rolü.

Tezi, Eskisehir, 2007. Fascin, hücre motilite ve adezyonunda önemli bir rol oynayan
aktin baglayıcı proteindir. Son çalısmalar tümör invazyonunda fascin’in önemli bir
medyatör oldugunu göstermistir. Bu çalısmada invaziv meme karsinomlarında fascin
ekspresyonu ile hasta yası, tümör boyutu, tümör histolojik derecesi (grade), aksiller
lenf nodlarında metastaz varlıgı ve sayısı, östrojen ve progesteron reseptör ve c-erbB-
2 onkoproteini ekspresyonu arasındaki iliskiyi incelenmistir. Ayrıca tüm
parametrelerin kendi aralarındaki iliskiyi ortaya koymak üzere istatistiksel analizleri
yapılmıstır. Olguların özellikleri ve klinikopatolojik bulguları hastane kayıtlarından
ve Patoloji Anabilim Dalı arsivinin 1996-2002 yılları arasındaki raporlarından temin
edilmistir. Doku spesmenleri primer meme kanserli 100 kadın olgudan elde
edilmistir. WHO klasifikasyonuna göre bu tümörler histolojik olarak; invaziv duktal
karsinom (n=91, %91), mikst duktal-lobüler karsinom (n=4, %4), invaziv lobüler
karsinom (n=2, % 2), tübülolobüler karsinom (n=2, %2) ve tübüler karsinom (n=1,
%1) seklinde kategorize edilmistir. Fascin ekspresyonu bu tümörlerden 12’sinde
(%12) pozitif olarak izlenmistir. Fascin ekspresyonu ile tümör histolojik derecesi
arasında belirgin bir iliski (P<0.001) izlenmis olup, fascin ekspresyonu daha çok
yüksek tümör histolojik derecesine sahip tümörlerde saptanmıstır. Fascin pozitif
olguların 11’inde (%91,6) östrojen ve progesteron reseptörü negatifti. Fascin
ekspresyonu ile hormon reseptör negatifligi arasında belirgin bir korelasyon
izlenmistir (östrojen için P=0.002, progesteron için P=0.007). Çalısmamızda fascin
ekspresyonu ile hasta yası, tümör boyutu, lenf nodu metastazı, metastatik lenf nodu
sayısı ve c-erbB-2 onkoprotein ekspresyonu arasında belirgin bir korelasyon
bulunmamıstır (P>0.05).
The role of fascin in the progression of invasive breast

protein that plays an important role in cell motility and adhesion. Current studies
showed that fascin is an important mediator in tumor invasion. In this study we
analysed the relation between the expression of fascin and age of case, tumor size,
tumor grade, axillary lymph nodes status and numbers , oestrogen and progesterone
receptor status and c-erbB-2 oncoprotein in invasive breast carcinomas. In addition,
the relation among all these parameters were analyzed statistically. Patient
characteristics and clinicopathologic finding were obtained from hospital records and
from the Pathology Department files between 1996 and 2002. Tissue specimens were
obtained from 100 female patients with primary invasive breast cancer. The tumors
were histologically categorized as invasive ductal carcinomas (n=91, %91), mixt
ductal-lobular carcinomas (n=4, %4), invasive lobular carcinomas (n=2, %2),
tubulolobular carcinomas (n=2, %2) and tubular carcinoma (n=1, %1) according to
the World Health Organization classification. Twelve of these tumors (%12) were
positive for fascin expression. Fascin expression correlated significantly with tumor
grade (P<0.001). Oestrogen and progesterone receptors were negative in the eleven
of fascin positive cases (%91,6). Fascin expression showed a remarkable correlation
with hormone receptor negativity (for oestrogen P=0.002, for progesterone P=0.007).
In our study no significant correlation was found between fascin expression and age
of case, tumor size, lymph nodes metastasis, numbers of lymph nodes involved and
c-erbB-2 oncoprotein expression (P>0.05).
