Erişkin sıçanlarda etan dimetan sülfonat tarafından oluşturulan testis
hasarının ve bu hasarın önlenmesinde çeşitli vitaminlerin etkinliğinin farklı

Çalışmamızda tek doz EDS uygulaması sonucunda sıçan testisinde görülen hasarın farklı
doz ve sürelerde E vitamini ve C vitamini kullanılarak ne oranda geriye döndürülebildiğini
araştırmayı amaçladık. Deneyde her birinde 16 adet erişkin erkek Sprague-Dawley cinsi
sıçan bulunan 5 grup oluşturuldu. Grupların, 1-EDS 2-DMSO (dimetilsülfoksit) 3-EDS+E vit,
4-EDS+C vit, 5-EDS+E vit+C vit şeklinde olması planlandı. Her gruptaki 16 hayvan eşit
sayıdaki iki alt gruba ayrıldı (enjeksiyon sonrası 7 gün ve 14 gün olmak üzere). Deney öncesi
her hayvanın vücut ağırlıkları tek tek tartıldı. EDS ilgili gruplara 75 mg/kg tek doz i.p.olarak,
1:3 oranında dimetilsülfoksit (DMSO) ile saf su içinde çözündürülerek deneyin ilk gününde
verildi. E vit 100 mg/kg, C vit ise 200 mg/kg i.p. olarak deney süresi bitene kadar günde tek
doz olarak verildi. Gruplardaki hayvanların yarısı 7. gün tartı sonrası anestezik madde
verilmesinin ardından (Ketamine+Xylazine 90+10 mg/kg i.p.) derhal servikal dislokasyon ile
öldürüldü. Geri kalan hayvanlar ise 14. gün sonunda aynı işlemlerle öldürüldü. Her iki testis
çıkarılıp tartıldıktan sonra PAS+Hematoksilin boyama için sol testis Bouin fiksatifi içine sağ
testis ise, immünohistokimyasal boyama için nötral tamponlu formaldehit içine alındı. Rutin
testis takibi sonrası bloklanan örneklerden 5μm kalınlıkta kesitler alındıktan sonra
PAS+Hematoksilinle boyanarak histopatolojik inceleme yapıldı. Serumda hormon miktarının
tayini için gereken kan, göğüs kafesi açılarak doğrudan kalpten alındı. Sonuçta EDS sonrası
oluşan testis ağırlığındaki azalma, E ve C vitaminlerinin gerek tek tek, gerekse beraber
verilmeleri ile normale döndürülemedi. Ancak E ve C vitaminlerinin histopatolojik skor, serum
testosteron seviyesi ve interstisyel Leydig hücreleri üzerine olumlu etki gösterdiği görüldü.

We aimed in this study to investigate the potential recovery effects of vitamins C and E in
different doses and two different time intervals against ethane dimethane sulfonate (EDS)-
induced testicular degenerations. Animal groups that planned to use in this study and their
treatments are as follows: Group I: Each animal received a single i.p. dose of EDS, Group II:
Each animal received a single i.p. dose of vehicle (dimethylsulfoxide and bidistilled water at
1:3 ratio), Group III: Each animal received a single i.p. dose of EDS, and subsequently daily
i.p. injections of vit E, Group IV: Each animal received a single i.p. dose of EDS, and
subsequently daily i.p. injections of vit C, Group V: Each animal received a single i.p. dose of
EDS, and subsequently daily i.p. injections of vit C+vit E. All groups of the rats subdivided
into two subgroups that include 7 rats in each and killed after 7 or 14 days after first
injections. The doses that we use were 75 mg/kg bw for EDS, 100 mg/kg bw for vit E, and
200 mg/kg bw for vit C. The body weights (immediately before the start and at the end of the
study) and both testicular weights measured. Animals anaesthetized with a mixture of
ketamine and xylazine at the doses of 90 and 10 mg/kg bw respectively and then killed by
cervical dislocation. Testes processed for routine paraffin technique. Tissue blocks were cut
at a thickness of 5 μm and then these were stained either with PAS+hematoxyline method or
with immunohistochemistry. Testicular sections were evaluated by light microscopy. Serum
testosterone levels were measured. İn conclusion we observed that both vitamin E and C
have positive effects on histopathologic score, blood testosteron level and amounth of Leydig
cells in testis tissue.
