Son yıllarda malignansiler üzerine etkilerinin ortaya çıkmasından dolayı bor
bileşiklerine olan ilgi gittikçe artmaktadır. Bor bileşiklerinin kanser hücreleri üzerinde
proteozom inhibitörü olarak çalıştığı birçok çalışmada gösterilmiştir (13). Çalışmamızda
sağlıklı insan lenfositleri ve HL-60 (lösemi) hücreleri üzerinde borik asit ve sodyum tetra
borat (boraks) bileşiklerinin etkilerini ayrı ayrı MTT, Neutral Red, Akış hücremetre ve
Geçirimli elektron mikroskobunu kullanarak değerlendirdik. Sitotoksik etkilerini saptamak
için 100, 250, 500, 1000 μM konsantrasyonlarda hücrelerimize borik asit veya sodyum
tetra borat verdik ve bunun için MTT yöntemini kullanıldık. Borik asitte 500 μM’da % 50,
sodyum tetra boratta ise ölü hücre oranının % 40’ lara ulaştığını bulduk. Diğer taraftan
kontrol olarak kullandığımız sağlıklı insan lenfositlerinde bu oran en yüksek dozda bile %
20’ leri geçmemektedir. Lizozomal aktivite açısından etkileri görebilmek için de
hücrelerimizi nötral kırmızısı boyasıyla boyadık. Aynı konsantrasyonlarda her iki bileşikte
de % 10 kadar lizozomal aktivitede bir azalış saptadık. Sağlıklı insan lenfositlerinde ve
lösemi hücrelerinde borik asit ve sodyum tetra boratın apoptotik etkisini akridin oranj ile
akış hücremetrede değerlendirdik. Borik asitin 1000 μM olan konsantrasyonunda sağlıklı
lenfositlerde % 2,5, HL-60 hücrelerinde % 8,8 apoptotik etki bulduk, sodyum tetra boratta
ise 1000 μM da sağlıklı lenfositlerde % 3,1, HL-60 hücrelerinde ise % 4,9 apoptotik etki
bulduk. Bor bileşiklerinin akut lösemi hücreleri üzerindeki ince yapı değişikliklerini
saptayabilmek için transmission (geçirimli) elektron mikroskobunda inceledik. Borik asit
500 μM konsantrasyonunda hem akut lösemi hücrelerinde hem de lenfositlerde çift
nukleus ve mikronukleus oluşturdu. Akut lösemi hücrelerinde mitokondrilerin boyutlarında
büyüme, kristalarında deformasyonlar ortaya çıktığını saptadık.
Bizim sonuçlarımıza göre, sağlıklı lenfositlerle akut lösemi hücrelerini
kıyasladığımızda bor bileşikleri lösemi hücrelerine daha etkilidir ve bu toksik etkiyi
mitokondriler üzerinden göstermektedir.
Recently, as the effects of malignancies have come out, the attention on boron
compounds has been taken and is gradually increasing. There are experiments in which
boron compounds work as a proteosom inhibitor on cancer cells (13). In our study, we
investigated the effects of boric acid and sodium tetra borate on MTT, Neutral Red, Flow
cytometry, transmission electron microscope methods on acute leukemia cell lineage and
healthy human lymphocyte. We used MTT method to evaluate cytotoxic effect by
following the one night incubation of leukemic cells in 100, 250, 500, 1000 μM
concentrations with boric acid and sodium tetra borate. We found that in boric acid at 500
μM there were dead cells at the rate of % 50 and in sodium tetra borate at the rate of % 40.
On the other hand, in healthy human lymphocytes used as a controller this rate did not
reach % 20s even in the highest concentration. In terms of lysosomal activity to see the
effects we dyed the cells with the neutral red by following the one night incubation of boric
acid and sodium tetra borate in 100, 250, 500, 1000 μM concentrations. We detected a
decrease % 10 in both compounds in the same concentrations and in the lysosomal activity.
We evaluated the apoptotic effect of boric acid and sodium tetra borate to lymphocyte and
leukemic cell line by flow cytometry using acridine orange methods. Apoptotic effect was
found boric acid 1000 μM concentration % 2,5 in normal lymphocyte, 1000 μM
concentration % 8,8 HL-60 cell lines and sodium tetra borate 1000 μM concentration %
3,1 normal lymphocyte, 1000 μM concentration % 4,9 HL-60 cell lines respectively. We
examined the boron compounds by means of transmission electron microscope method to
detect their effects on the acute leukemia cells. We observed that boric acid in 500 μM
concentration caused the double nucleus and micronucleus formation in both acute
leukemia cells and lymphocytes. We determined that an expanding in the dimension of
mitochondrium and a deformation in cristas appeared.
According to our results, boron compounds show a toxic effect on the acute
leukemia cells with comparing normal lymphocyte and this effect is via with the
mitochondrial pathway.
