The present study was aimed to investigate the protective effects of selenium
(Se) on cyclophosphamide (CP)-induced renal toxicity in rats. Eighty-four Sprague Dawley rats were equally divided into twelve groups of seven rats in each (control, 50
CP, 100 CP, 150 CP, 0.5 Se, 1 Se, 50+0.5 mg/kg CP+Se, 100+0.5 mg/kg CP+Se,
150+0.5 mg/kg CP+Se, 50+1 mg/kg CP+Se, 100+1 mg/kg CP+Se and 150+1 mg/kg
CP+Se).
The degree of protection produced by Se was evaluated by determining the level
of malondialdehyde (MDA) and the activity of endogenous antioxidant (GSH) were
estimated from kidney homogenates, and kidney were histologically examined as well.
Se elicited significant kidney protective activity by decreasing the level of lipid
peroxidation (MDA) and elevating the activity of GSH. Furthermore, these biochemical
observations were supported by histological findings.
In accordance with increasing doses of CP (50, 100 and 150 mg/kg) there was
toxicity in the kidney. When given together with the above CP doses, 0.5 and 1 mg/kg
Se considerably reduced the CP related toxicity in the kidney. When compared to 0.5
and 1 mg/kg Se in 50 and 150 mg/kg CP groups, 0.5 mg/kg Se was more protective on
50 mg/kg CP-induced toxicity while 1 mg/kg Se was more protective on 150 mg/kg CP induced renal toxicity.
Our data suggest that well-adjusted selenium dose may result in more
therapeutic effect against increased cyclophosphamide doses.
Bu çalışmada sıçanlarda siklofosfamid (CP)’ in neden olduğu renal toksisite
üzerinde selenyumun (Se) koruyucu etkisinin olup olmadığını araştırmak amaçlandı.
Sprague-Dawley cinsi 84 sıçan her grupta 7 hayvan olacak şekilde 12 gruba bölündü
(kontrol, 50 CP, 100 CP, 150 CP, 0.5 Se, 1 Se, 50+0.5 mg/kg CP+Se, 100+0.5 mg/kg
CP+Se, 150+0.5 mg/kg CP+Se, 50+1 mg/kg CP+Se, 100+1 mg/kg CP+Se ve 150+1
mg/kg CP+Se).
Se’ un böbrek homojenatlarındaki koruyuculuk derecesi lipit peroksidasyon
ürünü malondialdehid (MDA) ve endojen antioksidan olan glutatyon (GSH) düzeyi
belirlenerek değerlendirildi. Böbrekler histolojik olarak da incelendi. Se böbrekte lipit
peroksidasyonunu (MDA) azaltarak ve GSH aktivitesini artırarak anlamlı derecede
koruma sağladı. Bu biyokimyasal gözlemler histolojik bulgular tarafından da
desteklendi.
CP dozunun artışına paralel olarak (50, 100 ve 150 mg/kg) böbrekte hasarlar
saptandı. Yukarıdaki CP dozlarıyla birlikte verildiğinde, 0.5 ve 1 mg/kg Se böbrekte
CP nedenli toksisiteyi büyük oranda düşürdü. 50 ve 150 mg/kg CP verilen gruplar 0.5
ve 1 mg/kg Se dozları bakımından karşılaştırıldığında; 0.5 mg/kg Se 50 mg/kg CP
toksisitesini, 1 mg/kg Se ise 150 mg/kg CP toksisitesini azaltmada daha etkili oldu.
Verilerimiz, selenyum dozunun belirli oranlarda değiştirilmesiyle, artan CP
dozuna karşı daha güçlü bir koruyucu etkinliğin sağlanabileceğini düşündürmektedir.
