Bu çalısmada, Türk populasyonunda VDR geni Bsm I ve Fok I
polimorfizmleri ile prostat kanseri arasındaki iliskiyi arastırmak amaçlanmıstır.
Prostat kanserli 75 hasta ve benign prostat hiperplazili 112 kontrolün
periferik kanından elde edilen DNA’larda Bsm I ve Fok I lokusları PCR ile çogaltılmıs,
ardından PCR ürünleri Bsm I ve Fok I restriksiyon enzimleri ile kesilmis ve agaroz jel
elektroforezi ile analiz edilmistir.
Yapılan çalısma sonucunda, VDR Fok I genotip frekansları hastalarda
%48.0 FF, %41.3 Ff ve % 10.7 ff; kontrollerde % 41.1 FF, % 48.2 Ff ve %10.7 ff olarak
saptanmıstır. F allel frekansı hastalarda % 89.3, kontrollerde ise %89.2 olarak
bulunmustur. f allel frekansı hastalarda %52.0, kontrollerde %58.9 olarak saptanmıstır.
VDR Bsm I frekansları hastalarda % 22.6 BB, % 41.3 Bb, % 36.0 bb;
kontrollerde %33.9 BB, % 42 Bb ve % 24.1 bb olarak saptanmıstır. B allel frekansı
hastalarda %64.0, kontrollerde % 75.9 olarak bulunmustur. b allel frekansı hastalarda %
77.3, kontrollerde % 66.1 olarak saptanmıstır.
Sonuç olarak, Türk populasyonunda Fok I ve Bsm I genotip dagılımlarının
prostat kanseri hastaları ve kontrollerde istatistiksel olarak önemli bir farklılık
göstermedigi (P> 0.05), dolayısıyla bu polimorfizmlerin prostat kanseri tanısında
genetik markır olarak kullanılmaya uygun olmadıkları görülmüstür. Hastaların
histopatolojik bulgulara göre gruplandırılması da sonucu degistirmemistir.
In this study, we aimed to investigate the association between VDR gene
Bsm I and Fok I polymorphisms with prostate cancer in Turkish population.
In the DNA’s extracted from pheripheral blood of 75 prostate cancer cases
and 112 benign prostatic hyperplasia controls, Bsm I and Fok I locuses were amplified
by PCR. Subsequently, PCR products have been digested by Bsm I and Fok I restriction
enzymes and results were analysed by agarose gel electrophoresis.
As result, VDR gene Fok I genotype frequencies were 48.0%, FF, 41.3 %,
Ff, 10.7 % ff in the cases; 41.1 %, FF; 48.2 %, Ff; and 10.7 %, ff in the controls. The
frequencies of F allels in the cases were 89.3 %, and were 89.2 % in the controls. The
frequencies of f allels were 52.0 % in the cases and were 58.9 % in the controls.
VDR gene Bsm I genotype frequencies were 22.6 %, BB, 41.3, Bb, 36.0
% bb in the cases; 33.9 %, BB; 42.0 %, Bb; and 24.1 %, bb in the controls. The
frequencies of B allels in the cases were 64.0 %, and were 75.9 % in the controls. The
frequencies of b allels were 77.3 % in the cases and were 66.1 % in the controls.
In conclusion, Fok I and Bsm I genotype distribution between prostate
cancer cases and controls were not significantly different (P>0.05) in Turkish
population. So, according to this study, we conclude that these polymorphisms are not
usefull as markers of prostate cancer. Stratification of cases and according to
pathological findings did not changed the results.
