İlaç adayı moleküllerin belirlenmesine yönelik çalışmalar üç ana grupta toplanmaktadır. Bunlar, kara bitkileri, kara hayvanları, deniz organizmaları ve mikroorganizmaların metabolitlerinin oluşturduğu doğal kaynaklı bileşikleri temel alan çalışmalar, enzim-substrat veya reseptör teorisine dayalı “kombinatoryal kimya” teknolojisi ile üretilen ve sayıları milyonları bulan sentetik kimyasal maddeleri temel alan çalışmalar ve rekombinant teknolojileri ile üretilen biyoteknolojik ilaçlardır. Biyolojik etki gösteren bileşikler, doğrudan ilaç etken maddesi veya yarı sentezle değiştirilerek daha etkili ve daha az toksik moleküller haline dönüştürülmek suretiyle kullanılabilmektedir.
Bu tez projesi çalışmasında farklı biyolojik aktivite gösterdiği bilinen abiyetik asit, betulin, hidroksi diizoforon ve oleik asit moleküllerinin biyotransformasyon tepkimeleri ile türevlerinin elde edilmesi ve biyolojik aktivitelerinin ortaya konması amaçlandı. Ön biyotransformasyon tepkimeleri için 28 farklı mikroorganizma kullanıldı. Betulin ile hidroksi diizoforon kullanılarak gerçekleştirilen ön biyotransformasyon tepkimelerinden yeni metabolit elde edilemedi. Abiyetik asitin Mucor ramannianus ve Neurospora crassa ile biyotransformasyon tepkimeleri sonucu monohidroksillenmiş üç farklı türevi olan [2α-, 7β- ile 1β-hidroksi dehidroabiyetik asit] sırasıyla %12.7, 15.5 ile 20.2 verimle elde edildi. Oleik asit biyotransformasyonu sonucu çok sayıda metabolit içeren karışımlar elde edildi. In vitro antimikrobiyal aktivite deneyleri broth mikrodilüsyon yöntemi kullanılarak 19 farklı patojen mikroorganizmaya karşı gerçekleştirildi. Abiyetik asit ve türevlerinin izolatlara karşı test edilen bütün derişimlerde önemli bir aktivite gözlemlenmedi (>250 μg/mL). A549 ve HepG2 kanser hücreleri kullanılarak gerçekleştirilen sitotoksisite çalışmaları sonucunda ise 7β-hidroksi dehidroabiyetik asit türevinin HepG2 (İK50=196.6 μg/mL ve SI>187) hücrelerine karşı en etkili ve seçici antikanser etkiye sahip olduğu tespit edildi.
Studies aimed at determining a drug candidate molecule can be classified into three groups. These are; studies based on naturally occurring chemical substances obtained from land plants, land animals, sea organisms and microorganisms’ metabolites, studies based on synthetic chemical compounds generated through combinatorial chemistry technology, approaching to millions numerically, by using enzyme-substrate interactions or reseptor theory and biotechnological medicines produced via recombinant technology. Naturally occurring compounds demonstrating biological activity can be used either directly as active ingredient for medicines or they are utilized after making them less toxic and more potent active ingredient by substituting them with their semi-synthesis.
In this study we aimed at obtaining derivatives of abietic acid, betulin, hydroxy diisophorone and oleic acid which are known to exhibit a variety of biological properties through bioconversion reactions and investigating the biological activities of the metabolites. 28 different microorganisms were used for pre-biotransformation experiments. No metabolites were obtained for betulin and hydroxy diisophorone biotransformations. However, biotransformation reactions of abietic acid by Mucor ramannianus and Neurospora crassa produced three monohydroxylated derivatives [2α, 7β and 1β-hydroxy dehydroabietic acid] with 12.7, 15.5 and 20.1% yields, respectively. Biotransformation reactions of oleic acid by fungi produced mixtures containing a large number of metabolites. In vitro antimicrobial activity studies were carried out against 19 different pathogen microorganisms via using the broth microdilution methods. No significant activity was observed for microorganisms at all tested concentrations of abietic acid and its derivatives (>250 μg/mL). According to cytotoxicity studies using A549 and HepG2 cancer cells, 7β-hydroxy dehydroabietic acid derivative was found to be the most effective and selective anticancer effect against HepG2 cell line (IC50=196,6 μg/mL and SI>187).
