Çörek otu ve nane yağı yurdumuzda çeşitli amaçlarla kullanılmaktadır.
Rinosinüzit ve alerjik rinit tedavisi bu kullanım alanları arasındadır. Bu çalışmada,
çörek otu ve tıbbi nane uçucu yağının inhalasyon yolu ile sinüs mukozasına ulaşım
miktarının araştırılması hedeflendi. Toplam 18 adet rat, her grupta 6 adet rat olacak
şekilde randomize 3 gruba ayrıldı. Grup 1; çörek otu uçucu yağ, grup 2; nane uçucu
yağ grubu, grup 3; kontrol grubu olarak adlandırıldı. Çörek otu uçucu yağı; % 4
(h/h), nane uçucu yağı; % 1 (h/h) oranlarında kaynamış distile su ile karıştırılarak
20*20*25 santimetre ölçülerinde kapalı sistem cam fanus içerisine yerleştirildi. Grup
1 ve 2 kapalı sistem cam fanus içerisinde uçucu yağa 20’şer dakika inhalasyona
maruz bırakıldı. Ratlar dekapite edilerek sinüs diseksiyonu yapıldı. Cam örneklem
şisesi içerisindeki sinüs mukozasından uçucu bileşenlerin adsorpsiyonu için Tepe
Boşluğu Katı Faz Mikroekstraksiyon (TB-KFME) yöntemi kullanıldı. Adsorbe
edilen uçucu yağ bileşenleri GK-KS yöntemi ile tespit edildi. Çörek otu uçucu yağı
inhalasyonuna maruz bırakılan ratların sinüs mukozasında α-pinen temel bileşen
olarak tespit edildi. Nane uçucu yağı inhalasyonuna maruz bırakılan sıçanların sinüs
mukozasında 1,8- sineol temel bileşen olarak tespit edildi. Çörek otu ve nane uçucu
yağının inhalasyon yolu ile kullanımında sinüs mukozasına ulaşım miktarının
araştırılmasını amaçladığımız bu çalışmada, inhalasyon yolu ile kullanıldığında, her
iki uçucu yağın, sinüs mukozasına ulaştığı gösterilmiştir. Topikal etkinliği bir çok
kinik araştırmada gösterilmiş ve yıllardır tedavi alanında güvenle kullanılan ilaçlara
benzer şekilde, antienfektif, antioksidan ve antiallerjik etkinliği gösterilmiş olan
çörek otu ve nane uçucu yağının da topikal uygulama ile kullanımının tedavide
başarılı sonuçlar vereceği kanaatindeyiz. Çalışmamızın bu konudaki yapılacak
farmakolojik, toksikolojik ve sonrasında klinik çalışmalar için de öncülük
edebileceği kanaatindeyiz.
Cumin and peppermint essential oil were used for various purposes in our
country. Treatments of rhinosinusitis and allergic rhinitis are among these purposes.
In this study, we aimed to investigate the amount of black cumin and peppermint
essential oil that can reach to sinusal mucosa via inhalation. Eighteen rats were
randomized into 3 groups that contain 6 rats in each group. The first group; black
cumin essential oil, the 2nd group; peppermint essentai oil and the 3rd group is named
as control group. Cumin essentail oil is mixed with distilled water in the ratio of 4%
(h/h) and peppermint essential oil is mixed with distilled water in the ratio of 1%
(h/h). Afterwards they were stored in a closed glass lantern in the size of 20*20*25
cm. First and the 2nd groups were exposed to volatile oils in the aforementioned
glass lanterns for 20 minutes. Afterwards rats were decapitated and subsequently
sinus dissections were performed. Solid Phase Microextraction (SPME) method was
implemented for adsorption of essential components stems from the sinusal mucosa
in the glass sampling bottle. The essential oil components were detected by GK-KS
method. α-pinen was detected as the main component in the sinusal mucosa of the
rats that were exposed to cumin essential oil inhalation. 1,8- sineol was detected as
the main component in the sinusal mucosa of rathat that were exposed to peppermint
essential oil inhalation. It is shown that both of the essential oils reach to sinusal
mucosa via inhalation in this study that we aimed to investigate the amount of cumin
and peppermint essential oils via inhalation. In conclusion, cumin and peppermint
essential oils have been already shown to be anti-infectious, antioxidant and
antiallergic similar to the medications that have been already found to be topically
effective in many clinical studies and safely implemented in treatment for years.
Therefore, we think that their topical application might yield successful therapeutic
results. It is expected that our study would facilitate further pharmacology,
toxicology and clinicial studies.
