Meme kanseri kadınlar arasında en sık görülen kanser türüdür. Hayati önem taşıyan
organlarda gelişen sekonder tümörler, meme kanserinden kaynaklı ölümün en önemli
nedenini oluşturmaktadır. Bu yüzden, metastaz gelişiminin önlenmesinin ölüm oranını
büyük ölçüde düşüreceği düşünülmektedir. Günümüzde meme kanseri tanısında ve
takibinde biyobelirteçler kullanılmasına karşın, metastaz gelişiminin belirlenmesinde yeni
biyobelirteçlere ihtiyaç duyulmaktadır. Bu belirteç, invaziv olmayan yollardan kolaylıkla
elde edilmeli, hastanın prognozu hakkında bilgi vermesinin yanında, tümör dokusuna
spesifik mutasyonlar hakkında yorum yapılmasına olanak sağlamalıdır.
Serbest DNA’lar, plazma ve serumda bulunan tümör hücrelerine ait DNA fragmentleridir.
Yapılan çalışmalar serbest DNA’ların kanser hastalarında prognostik ve diagnostik açıdan
değere sahip olduğunu göstermektedir. Serbest DNA’ların, hassas hücreleri etkileyerek,
metastaza neden olabileceği çeşitli hayvan modeli çalışmalarında gösterilmiştir. Gen
transferi ile gerçekleştiği düşünülen bu olay, daha sonra “Horizontal Gen Transferi” olarak
adlandırılmış olup “Genometastaz Hipotezi” olarak terminolojiye geçirilmiştir
Yapılan bu çalışmada, ko-kültür sisteminde MDA-MB-231 meme kanser hücreleri ile
sağlıklı osteoblast hFOB 1.19 hücrelerindeki APC, RASSF1A ve p53 genleri için horizontal
gen transferi MSP/PCR yöntemi ile araştırılmıştır. Ayrıca DNaz I’in gen transfeksiyonu
üzerine olan etkileri de incelenmiştir. Muhtemel gen transferi ile hücrelerin canlılık,
proliferasyon ve adezyon özellikleri üzerine etkisi Tripan blue ve XTT yöntemleri esas
alınarak belirlenmiştir.
Çalışma sonucunda, metile APC1A, APC1B ve p53 ekson 8 horizontal gen
transfeksiyonunun hFOB 1.19 hücrelerine gerçekleştiği belirlenmiştir. Bu transfeksiyon
sonucunda, hFOB 1.19 hücrelerinin adeziv özelliklerinde değişim olduğu gözlenmiştir.
DNaz I’ in hücrelerde transfeksiyonu hızlandırdığı analiz edilmiştir.
Breast cancer is the most common and deadly cancer type in women. The main cause of
death the secondary tumor developing to vital organ that appear at the end of the process of
metastasis. Therefore, its believed that prevention of metastasis development also
significantly reduce the mortality. Today however there are biomarkers for breast cancer
diagnosis, there are also need to develop new biomarkers for metastasis detection for early
period. The biomarker must be readily obtained from non-invasive ways and it should
allow to comment on specific mutations in the tumor tissue and provide information about
the patient's prognosis. Cell free DNA’s are tumour cell derived DNA fragments which are
present in plasma and serum. It has been shown in many studies that cell free DNA may
have diagnostic and prognostic value for cancer patients. It has been shown in animal
studies that tumour cell derived-cell free DNA may affect sensitive cell and can cause
metastasis. This event is thought to occur by gene transfer that named as “Genometastasis
Hypothesis” and this gene transfer termed as “ Horizontal Gene Transfer”. In this study,
we cultured with MDA-MB-231 breast cancer cell line and hFOB 1.19 osteoblast cell line
in co-culture system and then investigated APC, RASSF1 and p53 gene horizontal gene
transfer with MSP/PCR that investigated DNase I affect on gene transfection in the coculture
system. And the determined the cell proliferation, adhesion and viability values
with Tripan blue and XTT. As a result of the study, we identified methylated APC1A,
APC1B and p53 exon 8 horizontal gene transfer MDA-MB-231 cells to hFOB 1.19 cells.
Result of the transfection, we demonstrated that result of the horizontal gene transfection
hFOB 1.19 cells gained malignant cell adhesive properties and the transfection was
increased by DNase I.
