Taze fasulye (Phaseolus vulgaris L.) genotiplerinin yüksek sıcaklığa toleranslarında genotipik farklılığın belirlenmesi amacıyla fizyolojik ve biyokimyasal parametrelerden yararlanılan bu çalışmada Balkız, Ferasetsiz ve Yerel Genotip kullanılmıştır.
Fideler, 3–4 gerçek yapraklı oluncaya kadar serada ortalama 15/30ºC (gece/gündüz) sıcaklıkta, ~%65 nemde yetiştirilmiştir. Bitkiler yüksek sıcaklık stresi denemeleri için laboratuardaki iklimlendirme kabinine alınarak, kabin sıcaklığı kademeli olarak 35, 40,45 ve 50ºC’ye yükseltilmiş ve her bir sıcaklık derecesinde 24 saat tutulmuştur. Her bir sıcaklık kademesindeki bitkilerden alınan yaprak örneklerinde, yaprak oransal sukapsamı (YOSK), turgor kaybı (TK), yaprak alanı, klorofil miktarı (KM) ve lipid peroksidasyonu (MDA) ölçümleri yapılmıştır. Ayrıca, SDS-PAGE yöntemiyle toplam çözünebilir protein profilleri çıkarılmıştır. YOSK sonuçlarına göre Ferasetsiz ve Yerel Genotip en yüksek YOSK’na sahip iken Balkız en düşük değere sahip olmuştur. Buna karşılık TK ve MDA miktarında Ferasetsiz ve Yerel Genotip en düşük değerlere sahipken Balkız en yüksek TK ve MDA miktarına sahip genotip olmuştur. Yaprak alanı ve KM ise genotiplerin sıcaklık toleransları ile ilişkili bulunmamıştır. SDS-PAGE'de uygulamalara göre değişen pek çok protein bandı görülmüştür.
Taze fasulye genotiplerinin yüksek sıcaklık toleranslarını belirlemek için, serada yetiştirilen bitkilerden alınan yaprak örnekleri, kontrollü olarak yüksek sıcaklık uygulamasının yapıldığı sıcak su banyosunda 60ºC’ye kadar kademeli olarak tutulmuşve iyon sızıntısı testi sonucuna göre her bir genotipe ait yüksek sıcaklık tolerans dereceleri belirlenmiştir. Eldeedilen sonuçlara göre genotiplerin sıcaklık stres tolerans noktaları Balkız genotipinde 45,5 °C, Ferasetsiz ve Yerel Genotipte ise sırasıyla 50,8 ve 49,9 °C olarak belirlenmiştir. Sonuç olarak, Ferasetsiz ve Yerel Genotip'in yüksek sıcaklığa tolerant, Balkız genotipinin ise daha hassas olduğu belirlenmiştir.
Physiological and biochemical parameters were used to investigate the effect of genotypic variation on the tolerance to high temperature in green bean (Phaseolus vulgaris L.) genotypes, (Balkız, Ferasetsiz, Yerel Genotip).
The seedlings were grown until 3-4 leaves stages under controlled conditions at 30/15ºC (day/night) temperature with RH ~%65 then transferred to climate chamber to expose to heat stress. The temperature was increased stepwise to 35, 40, 45 and 50ºC for 24 hours. Leaf relative water content (LRWC), loss of turgidity (LT), leaf area, chlorophyll content (CC) and lipid peroxidation (MDA) were measured in the leaves collected from the plants in each temperature step. Moreover, total soluble protein profiles were determined using SDS-PAGE method. According to LRWC results, Ferasetsiz and Yerel Genotype have the highest LRWC where the Balkız has the lowest value. On the other hand, Ferasetsiz and Yerel genotype have the lowest LT and MDA but Balkız has the highest. The leaf area and CC were not associated with the heat stress tolerance (HST) of genotypes. On SDS-PAGE many protein bands were detected depending on the treatments.
To determine HST of green bean genotypes, the leaf samples, collected from greenhouse, were kept gradually until 60ºC. Each green bean genotype’s heat stress tolerances was determined by ion leakage test results. According to results, HST was determined as 45,5 °C, 50,8 °C, 49,9 °C for Balkız, Ferasetsiz, Yerel Genotype respectively. As a result, Ferasetsiz and Yerel Genotype were heat tolerant genotypes, whereas Balkız was determined as more sensitive genotypes.
