Bu çalışmanın amacı naproksen sodyum (NapNa) etken maddesinin partikül
boyutuna ultrases şiddeti ve süresinin etkisi zeta potansiyel, UV, taramalı elektron
mikroskobu (SEM), X-ışınları kırınımı (XRD), fourier dönüşüm kızılötesi spektroskopisi
(FT-IR) ve diferansiyel tarama kalorimetri (DSC) ölçümleri kullanılarak araştırılmasıdır.
Farklı ultrases şiddeti ve süresinin ilaç etken maddelerinin yüzey morfolojisinde ve kristal
yapısında değişikliğe sebep olduğu belirlenmiştir. Ultrases uygulama süresini ve genliği ile
yüzey alan artırımı ve kristal boyuttaki küçülme ile lineer bir ilişki olmadığı bunun yerine
optimum ultrases süresi ve şiddetinin etken maddelerinin partikül boyutunu küçültmede,
yüzey alanını ve dolayısı ile çözünürlüğü artırmada oldukça etkin olduğu gözlenmiştir.
Çekirdeklenme, polimorf oluşumu ve istenen polimorf yapıyı elde etmek için optimum
ultrases uygulanma şiddeti ve süresi belirlenmiş ve etken maddenin uygun polimorfik
yapısının elde edilmesi için önemli bilgiler elde edilmiştir. Bulunan sonuçlara göre değişik
şiddet ve sürelerde ultrases uygulanmasının, polimorfik kontrol, eleme ve uygun
polimorfik yapı elde etmede büyük potansiyele sahip olduğu tespit edilmiştir. Polimorfik
kontrol, eleme ve uygun yapı elde etme yeni ilaç geliştirme ve keşfi için oldukça
önemlidir.
The purpose of this study is, to investigate the effect of ultrasound intensity and
expose time to the particle size of naproxen sodium (NapNa) active ingredient by using
zeta potential, UV, scanning electron microscope (SEM), X-ray diffraction (XRD),
Fourier transform infrared spectroscopy (FT-IR) and differential scanning calorimetry
(DSC) measurements. It has been determined that, different ultrasound intensity
and expose time duration has changed the surface morphology and crystal structure of the
drug active substances. It has been observed that, ultrasonic intensity and duration doesn’t
have a linear correlation with the increase on the surface area and decrease on the particle
size. On the contrary, it has been detected that optimum ultrasound intensity and duration
has a great effect on decreasing the particle size of drug active substances and increasing
the surface area, therefore increasing the solubility.
In this study, optimum ultrasound intensity and expose time values have been
detected to acquire the nucleation, polymorph formation and desired polymorph structure.
According to the obtained results, different ultrasound intensity and duration application
has a great potential to acquire polymorphic control, elimination and appropriate
polymorphic formation. The polymorphic control, elimination and obtaining appropriate
polymorphic formation are very important on discovery and development of new drugs.
