Bu çalışmada, göz damlası olarak kullanılan bazı ilaçların etken maddesi olan Olopatadin HCl (OLO) 'nin kapiler zon elektroforez (CZE) ile tayini için hassas ve seçici ayırma koşulları araştırılmıştır.
Tampon derişimi, tampona eklenen organik çözücülerin miktarı, pH, tamponun viskozitesi, kolon sıcaklığı, enjeksiyon hacmi, enjeksiyon süresi optimize edilmiştir. Analizler 20 mM, %15 (h/h) asetonitril içeren pH= 9,5 sodyum tetraborat tamponu kullanılarak, 50mbar’da 10 s enjeksiyon ile, 205 nm dalgaboyunda, 30 oC’da ve 25 kV potansiyel uygulanarak yapılmıştır. Uygun koşullar belirlendikten sonra yöntemin hassasiyetini ve tekrarlanabilirliğini arttırmak için uygun bir iç standart (IS) belirlenmiştir. Sistem için uygun olan metil paraben IS olarak kullanılmıştır. Geliştirilen yöntemin doğruluk, kesinlik, doğrusallık, geri kazanım, tayin sınırı (LOQ) ve gözlenebilme sınırı (LOD) gibi validasyon parametreleri araştırılmıştır. Bu yöntem OLO’nun tayini için göz damlası ilacına başarıyla uygulanmıştır. Bu çalışma OLO’nun CZE ile tayini açısından özgün bir çalışmadır.
In this study, sensitive and selective separation conditions were investigated with the capillary electrophoresis for the determination of Olopatadine HCl (OLO) which is an active ingredient of some medicines used as an eye drops. Buffer concentration, the quantity of the organic solvents added to buffer, pH the viscosity of the buffer, column temperature injection volume, injection time were optimized. Analyses were made using a run buffer consisting of 20 mM sodium tetraborate buffer of 15% ACN (acetonitrile) with pH 9.5, applying 50 mbar of pressure s at 10 s, at 205 nm and at 30 oC of fixed temperature. Certain parameters were used to find a suitable IS increases the repeatability and sensitivity of the developed method. Methyl paraben was suitable IS for this system. Validation parameters such as precision, accuracy, linearity, recovery, detection and quantification limits (LOD and LOQ) of the developed method were investigated. This method was applied for the determination of OLO in the pharmaceutical preparation of an eye drop. To the best of our knowlodge, this study is the first application for the determination of OLO by CZE.
