Gıda kaynaklı hastalıklar, insan sağlığını her geçen gün daha fazla tehdit etmektedir. Biyofilm oluşumu, bu enfeksiyonların en önemli kaynaklarındandır. Biyofilm, mikroorganizmaların bir yüzeye yapışması ve kendi ürettikleri ekstrasellülar matriks içine gömülmesi sonucu oluşturdukları organize komüniteler olarak tanımlanabilir. Gıda temas yüzeylerine patojenik mikroorganizmaların yapışması ve ardından biyofilm oluşturması gıda güvenliği açısından tehlikelidir.
Salmonella türleri ve Listeria monocytogenes gıda yoluyla bulaşan en önemli patojenik bakterilerdendir. Her iki bakteri de canlı ve cansız yüzeyler üzerinde biyofilm oluşturabilmektedir. Biyofilm içindeki bakteriler, temizleme ve sanitasyon işlemlerine artan bir direnç gösterirler. Bu çalışmanın amaçları (i) çeşitli gıda örneklerinden izole edilen ve tanımlanan Salmonella spp. ve L. monocytogenes suşlarının biyofilm varlığını araştırmak ve (ii) güçlü biyofilm üreten Salmonella spp. ve L. monocytogenes suşlarının çeşitli endüstriyel yüzeyler üzerindeki adezyon ve gelişim yeteneklerini değerlendirmektir. Analiz için alınan 603 gıda örneği içinde 6 izolat L. monocytogenes ve 8 izolat Salmonella spp. olarak tanımlanmıştır. İzolatlar standart mikrobiyolojik yöntemlerle tanımlanmıştır. Biyofilm tespitleri mikrotiter plak metodu ve tüp metodu ile belirlenmiştir. Test edilen tüm Salmonella spp. ve L. monocytogenes suşları biyofilm üretmiş ancak L. monocytogenes suşlarında daha yüksek derecede biyofilm belirlenmiştir. Bu iki testin yanında, seçilen güçlü biyofilm üreten suşların altı farklı endüstriyel yüzey (mermer, granit, cam, tahta, plastik, çelik) üzerindeki adezyon ve biyofilm oluşumları da SEM ile tespit edilmiştir.
Ayrıca seçilen iki suşun biyofilm oluşum ve gelişimleri granit yüzeylerde ve beş inkübasyon periyodunda değerlendirilmiştir (2., 4., 6., 24. ve 48. saatler). Olgun biyofilm oluşumu 24 ve 48. saatlerde tespit edilmiştir. Granit, mermer, tahta ve cam yüzeyler, çelik ve plastik yüzeylere kıyasla daha yoğun biyofilm oluşturmuşlardır. Özellikle granit ve mermer yüzeyler biyofilm oluşumu için daha elverişli ortamlar olarak bulunmuşlardır.
Human health is more threatened by food borne diseases day by day. Biofilm production is one of the most important source these infections. Biofilm is defined as an organise community, characterized by cells which are irreversibly attached to substrate or with each other. An adherence of pathogenic microorganisms to food contact surfaces and biofilm formation is dangerous with respect to food security.
Salmonella spp. and Listeria monocytogenes are important pathogenic bacteria, which are transmitted by food. It is known that both microorganisms may produce biofilm on biotic or abiotic surfaces. Bacteria in biofilms exhibit enhanced resistance to cleaning and sanitation. The aims of this study were (i) to investigate the biofilm existing of Salmonella spp.and L. monocytogenesis isolated and identified from variety food samples, (ii) to evalute the adhesion and growth abilities on variety industrial surfaces of strong biofilm positive Salmonella spp. and L. monocytogenesis strains by scanning electron microscope. Among the 603 food samples taken for analysis, a total of 6 samples were confirmed positive L. monocytogenes; 8 samples were confirmed positive for Salmonella spp. Isolates were identified by standard microbiological procedures. Biofilm detection was evaluated by microtiter plate assay and tube method. All tested Salmonella spp. and L. monocytogenes strains produced biofilm but strains of L. monocytogenes exhibited a higher ability of biofilm formation. Concominantly with these two methods, adhesion and biofilm formation of selected strong biofilm producing strains to six different industrial surface (marble, granit, glass, wood, plastic, steel) was also assessed by SEM.
In addition, biofilm formation and development of selected two strains were also evaluated on granite surfaces and at five incubation periods (2th, 4th, 6th, 24th and 48th). Mature biofilm formation was determined after 24 and 48 hours. Granite, marble, wood and glass surfaces presented higher intensity of biofilm, compared to the steel and plastic surfaces. Especially granite and marble are the surfaces in which we found to be the most convenient for the biofilm formation.
