Bu çalışmada KOAH'lı hastalarda mannoz bağlayıcı lektin (MBL) gen polimorfizmi ile KOAH arasındaki ilişkinin incelenmesi, serum MBL düzeylerinin saptanması ve KOAH alevlenme sıklığı ile serum MBL düzeyleri arasındaki ilişkinin incelenmesi amaçlanmıştır. Bu çalışmaya 80 hasta ve 60 sağlıklı (30 sigara kullanıcısı, 30 sigara kullanıcısı olmayan) kontrol dahil edildi. Hasta bireyler GOLD tanımına uygun olarak KOAH tanısı alan ve son 6 hafta içinde akut eksaserbasyonda olmayan ve kanser, tüberküloz (enfeksiyöz nedenler) gibi ek akciğer patolojisi olmayan olgulardan seçildi. Sağlıklı bireyler ise herhangi bir bilinen akciğer ve diğer sistem patolojisi olmayan gruptan seçildi. KOAH ile MBL polimorfizmi arasındaki ilişki Polymerase Chain Reaction-Restriction Fragment Lenght Polymorphism (PCR-RFLP) yöntemi kullanılarak araştırılmıştır. Serum MBL düzeyi ise Human Mannose-Binding Lectin Gene Elisa kiti kullanılarak belirlenmiştir. Çalışmamızın sonucuna göre MBL2 kodon 54B polimorfizmi genotip dağılımları açısından KOAH hastaları ile sağlıklı bireyler arasında anlamlı fark bulundu (p=0.026). KOAH ve kontrol grubu ortalama serum MBL düzeyleri (ng/ml) arasında anlamlı fark bulundu (p=0.021). Ancak KOAH alevlenme sıklığı ile serum MBL düzeyleri arasında bir ilişki saptanmadı. Sonuç olarak KOAH'lı hastalarla MBL polimorfizmi arasında ilişki saptandı ve ortalama serum MBL düzeyleri KOAH grubunda sağlıklı bireylere göre anlamlı olarak daha düşük ortaya kondu.
In this study, we aimed to investigate the relationship between mannose-binding lectin (MBL) gene polymorphism and COPD in patients with COPD, to determine serum MBL levels and to investigate the relationship between serum MBL levels and COPD exacerbation frequency. 80 patients and 60 healthy controls (30 smokers and 30 non-smokers) were included in the study. The patients were selected from the cases diagnosed with COPD according to the GOLD definition and who did not have acute exacerbations in the last 6 weeks and additional pulmonary pathologies such as cancer, tuberculosis (infectious causes). Healthy individuals were selected from the group without any known lung or other system pathology. The relationship between COPD and MBL polymorphism was investigated using the Polymerase Chain Reaction-Restriction Fragment Lenght Polymorphism (PCR-RFLP) method. Serum MBL level was determined using Human Mannose-Binding Lectin Gene Elisa kit. According to the results of our study, there was a significant difference between COPD patients and healthy subjects in terms of MBL2 codon 54B polymorphism genotype distributions (p = 0.026). There was a significant difference in mean serum MBL levels (ng / ml) between COPD and control group (p = 0.021). However, there was no correlation between the frequency of exacerbation of COPD and serum MBL levels. As a result, there was a relationship between patients with COPD and MBL polymorphism and mean serum MBL levels were significantly lower in the COPD group than in healthy subjects.
