Çalışmamızda astaksantinin (AST) sepsiste
antienflamatuar etkinliğinin araştırılmasını ve antienflamatuar etkinliği kanıtlanmış
olan deksametazon (DEKS) ile karşılaştırılmasını amaçladık. Çalışmamız, etik kurul
onayı sonrasında Eskişehir Osmangazi Üniversitesi TICAM laboratuarında toplam
40 sıçan 5 gruba ayrılarak yapılmıştır. Birinci gruba sadece dimetilsülfoksit
(DMSO), ikinci gruba lipopolisakkarit (LPS), üçüncü gruba AST, dördüncü gruba eş
zamanlı olarak LPS ve AST, beşinci gruba eş zamanlı olarak LPS ve DEKS
verilmiştir. Yirmi dördüncü saatte anestezi altında laparotomi sonrası karaciğer
dokusu ve kan örnekleri alınmıştır. İstatistiksel değerlendirme için GrapPad Prism6
programı kullanıldı. DMSO grubuna göre AST ve DEKS+LPS gruplarında plazma
TNF-alfa düzeyleri anlamlı düşük bulunmuştur. DEKS+LPS grubundaki TNF-alfa
düzeyleri AST+LPS grubundaki değerlere göre anlamlı olarak daha fazla düşmüştür.
Dokudaki TNF-alfa düzeylerinde LPS grubunda DMSO grubuna göre anlamlı
yükseklik bulunmuştur, AST+LPS ve DEKS+LPS gruplarında ise LPS grubuna göre
anlamlı olarak düşüklük bulunmuştur; ancak DEKS+LPS grubu ve AST+LPS grubu
arasında anlamlı fark bulunmamıştır. Doku Nükleer Faktör-kappa B düzeyleri
AST+LPS ve DEX+LPS gruplarında anlamlı olarak düşük bulunmuştur. Ancak iki
grup arasında anlamlı bir fark bulunmamıştır. Karaciğer dokusundaki apoptotik
hücre yüzdesi AST+LPS ve DEKS+LPS gruplarında LPS grubuna göre belirgin
düşük bulunmuştur. Plazmada ve karaciğer dokusunda bakılan sitokin düzeylerinde
AST grubunda olumlu sonuçlar elde edilmiştir. Aynı zamanda deksametazon
uygulaması ile karşılaştırıldığında farklılıklar olmakla birlikte benzer ölçüm değerleri
de elde ettik. Apopitoz görülen hücre oranları değerlendirildiğinde anlamlı olarak
AST’in etkin olduğu sonucuna ulaştık. Sepsis patogenezinin kompleks bir süreç
olması nedeniyle iyi tasarlanmış daha fazla sayıda çalışma yapılması gerekmektedir.
We aimed to investigate antiinflamatory effect of astaxanthin (AST) in sepsis
and compare with dexamethasone (DEX) which antiinflamatory efficiency was
proved. After local ethics commitee permitted the study was carried out at TICAM
labarotory dividing 40 rats in five groups. In first group only dimetil sulphoksit
(DMSO) was injected, in second group lipopolysakkarid (LPS) was injected, in third
group AST was injected, in fourth group LPS and AST were injected, in fifth group
LPS and DEKS were injected simultaneous to rats. At twenty forth hour, liver tissue
samples were taken under anesthesia by laparotomy and blood samples were taken.
GrapPad Prism6 programme was used for statistical analysis. In AST and
DEKS+LPS groups plasma TNF-alfa levels were found significantly low comparing
with DMSO group. TNF-alfa levels of DEKS+LPS group decreased more
comparing with the AST+LPS group. Tissue TNF-alfa levels were found
significantly high in LPS group comparing with the DMSO group, in AST+LPS and
DEKS+LPS groups tissue TNF-alfa levels were found significanty low comparing
with the LPS group but no significant difference was found between DEKS+LPS
and AST+LPS groups. Tissue nuclear factor-kappa B levels were found
significantly low in AST+LPS and DEX+LPS groups. But no significant difference
found between two groups. Apoptotic cell percent of liver tissue was found
significantly low in AST+LPS and DEKS+LPS groups comparing with the LPS
group. Positive results of plasma and liver tissue cytocin levels were obtained in
AST group. By the way, comparing with the DEX group there were some
differences but we found similar resultsWhen the percent of cells which apopitosis
seen was assesed; we found out that AST was significantly effective. Because of the
complicated sepsis pathogenesis process, well designed more studies have to be
done.
